MIB/HERC2 domain profile





Description

The MIB/HERC2 domain is a conserved region of ~80 amino acids, which has been identified in:

  • Animal Mind bomb 1 and 2 (MIB1 and MIB2) E3 ubiquitin ligases. They contain a very similar domain organization including an amino terminus ZZ-type zinc finger (see <PDOC50135>) surrounded by two MIB/HERC2 domains, followed by two MIB specific repeats, multiple ankyrin repeats (see <PDOC50088>), and two or more Ring fingers (see <PDOC00449>) at their C-terminal ends. They partake in the Notch signaling pathway by ubiquitinating the Notch receptors Delta and Jagged prior to endocytosis [1,2,3,4].
  • Animal probable E3 ubiquitin-protein ligase HERC2 (EC 6.3.2.-). It contains additional motifs, including RCC1 repeats (see <PDOC00544>), a DOC domain (see <PDOC51284>), a carboxy-terminal HECT domain (see <PDOC50237>), and a ZZ-type zinc finger [5].

The profile we developed covers the entire MIB/HERC2 domain.
